What is Amino Acid Analysis (AAA)?
Amino Acid Analysis (AAA) is a powerful analytical technique used to determine the composition and quantity of amino acids in a sample. By extension, AAA is a valuable tool for quantifying peptides. AAA allows for the precise measurement of individual amino acids, enabling the determination of peptide concentrations with high accuracy.
AAA was first developed by Vincent du Vigneaud in the 1930s and was later refined by William Stein and Stanford Moore, who won the Nobel Prize in Chemistry in 1972 for their work on the technique. Stein and Moore's method involved a high-performance cation exchange separation followed by a post-column derivatization that allows sensitive detection of the amino acids. Today, modern AAA techniques like AccQ•Tag™ technology have replaced the older assays and have shown to be highly accurate and reproducible in experienced labs.

With the dramatic increase in stable isotope labeled (SIL) peptides for mass spectrometry, AAA is required to provide exact molar quantitation and/or the most accurate distribution of peptides into single use vials. SciTide utilizes AAA to provide accurate and precise measurement of peptide concentrations for "mass spectrometry ready" custom products or other quantitative peptide uses.
Gross Weight vs Net Peptide Content/Net Amounts
Peptides are measured and sold by gross weight, which includes water and counterions such as trifluoroacetate, acetate or chloride. The mass of water and counterions are included in a peptides gross weight. Amino Acid Analysis (AAA) allows for the accurate measurement of Net Peptide Content (NPC), which is the amount of peptide present in a sample after subtracting the weight of the water and counterions.
Net Peptide Content (NPC) is particularly important for quantification, where it will be used to accurately distribute the peptide into single use vials by net amounts. Eliminating the need for cumbersome and inaccurate weighing of peptides while in the hands of our customers.
